خلاصه ای از پروژه:

ویروس نقص ایمنی انسانی (HIV)، عامل بیماری ایدز، از خانواده لنتی‌ویروس‌ها منشعب شده است و با آلوده کردن افراد، عفونت مادام‌العمر ایجاد می‌کند. در صورت عدم درمان، اکثر افراد آلوده به HIV در عرض یک دهه به دلیل نقص سیستم ایمنی ناشی از HIV، به عفونت‌های فرصت‌طلبانه مرگبار مبتلا می‌شوند. ایدز به عنوان یکی از مهم‌ترین مشکلات بهداشت عمومی در سطح جهان شناخته می‌شود.

ویروس HIV دارای ساختاری با هسته کروی و استوانه‌ای، ژنوم RNA تک رشته‌ای، پروتئین‌های پوششی با تغییرات آنتی‌ژنی، آنزیم ترانس کریپتاز معکوس و پروتئاز است. این ویروس از غشای پلاسمایی سلول میزبان جوانه می‌زند و با استفاده از ترانس کریپتاز معکوس، DNA ویروسی را از RNA ژنومی می‌سازد. ویژگی‌های برجسته این ویروس شامل غیرتومورزا بودن، احتمال سیتوتوکسیک بودن، توانایی آلوده کردن سلول‌های سیستم ایمنی و ایجاد بیماری‌های مزمن پیشرونده است.

ژنوم HIV شامل ژن‌های gag، pro، pol و env است که برای تکثیر ویروس ضروری هستند. ژن‌های اضافی نیز در تنظیم بیان ویروسی و پاتوژنز بیماری نقش دارند. تنوع ژنتیکی بالایی در ویروس HIV وجود دارد که ناشی از نرخ بالای تکثیر ویروسی و خطای بالای آنزیم ترانس کریپتاز معکوس است. مناطق با بیشترین تفاوت، ژن env را شامل می‌شوند که در اتصال ویروس به مولکول CD4 و گیرنده‌های کمکی نقش دارد.

HIV یک رتروویروس از خانواده Retroviridae و جنس Lentivirus است و دو نوع متمایز HIV-1 و HIV-2 دارد. منشا HIV در انسان، عفونت‌های بین گونه‌ای توسط ویروس‌های میمونی در مناطق روستایی آفریقا و تماس مستقیم انسان با خون آلوده پستانداران است. ویروس از طریق اتصال به مولکول CD4 و گیرنده‌های کمکی وارد سلول می‌شود و پس از ورود، RNA ویروسی به DNA تبدیل شده و در DNA سلول میزبان ادغام می‌شود.

مسیر عفونت HIV شامل مراحل عفونت اولیه، انتشار ویروس به اندام‌های لنفاوی، کمون بالینی، افزایش بیان HIV، بیماری بالینی و مرگ است. ویژگی اصلی عفونت HIV، کاهش سلول‌های T کمک‌کننده است. مونوسیت‌ها و ماکروفاژها نقش مهمی در انتشار و پاتوژنز عفونت HIV دارند و می‌توانند به عنوان مخازن اصلی ویروس در بدن عمل کنند. علائم عفونت حاد HIV غیر اختصاصی هستند، در حالی که علائم ایدز شامل سرکوب سیستم ایمنی و بروز عفونت‌های فرصت‌طلبانه شدید یا نئوپلاسم‌های غیر معمول است.
